Microglia are resident immune cells of the central nervous system that act as brain-specific macrophages and also known to regulate innate functions astrocytes through secretory molecules. This communication plays an important role in brain homeostasis well neuropathologic disease. In this study, we aimed elucidate whether microglia could crosstalk induce microglial polarization proliferation, which can be further regulated under a microenvironment mimicking stroke. mixed glial culture showed increased survival proliferation were altered M2 microglia; CD11b?GFAP+ resulted approximately tenfold increase cell after reconstitution astrocytes. Furthermore, GM-CSF stimulated induced them become CCR7+ M1 microglia, have phenotype suppressed by anti-inflammatory cytokines such IL-4, IL-10, substance P. addition, co-culture A2 phenotype; they activated A1 TNF-? IFN-? stroke-mimicking condition. Altogether, polarization, possibly acquisition both converted inflammatory environment. study demonstrated polarized astrocytes, respectively, vitro provides with explore how may behave disease milieu vivo transplantation.
